چکیده

Background Allergic asthma is associated with T helper 2 (Th2) immune response to an allergen. IL-4 and IL-13, key cytokines involved in Th2 immunity, promote eosinophilic inflammation and B cell isotype switching to IgE. House Dust Mite (HDM) allergen has been shown to induce allergic asthma in a mouse model via Th2 immune response that drives eosinophilia and influx of effector T cells into lung. An IL-4Raplha antibody that blocks signaling of IL-4 and IL-13 through both Type I (IL-4Ralpha/ IL-2Rgamma) and Type II (IL-4Ralpha/IL-13R) receptors, and an IL-13Ra2-Fc fusion protein that prevents binding and signaling of IL-13 via Type II receptor, were both tested in the HDM-induced mouse model of asthma.

Vaccination against IL-33 Inhibits Airway Hyperresponsiveness and Inflammation in a House Dust Mite Model of Asthma

In several clinical and experimental studies IL-33 and its receptor have been found to play important roles in the development of asthma and allergic airway inflammation. We evaluated the effects of vaccination against IL-33 in a mouse model of airway inflammation induced by house dust mite (HDM) allergen. Balb/c mice received the IL-33 vaccine subcutaneously, followed by intranasal administrat...

Central role of immunoglobulin (Ig) E in the induction of lung eosinophil infiltration and T helper 2 cell cytokine production: inhibition by a non-anaphylactogenic anti-IgE antibody

Elevated levels of immunoglobulin (Ig) E are associated with bronchial asthma, a disease characterized by eosinophilic inflammation of the airways. Activation of antigen-specific T helper (Th) 2 cells in the lung with the subsequent release of interleukin (IL) 4 and IL-5 is believed to play an important role in the pathogenesis of this disease. In this study, we have used a non-anaphylactogenic...

Neutralization of TSLP Inhibits Airway Remodeling in a Murine Model of Allergic Asthma Induced by Chronic Exposure to House Dust Mite

Chronic allergic asthma is characterized by Th2-typed inflammation, and contributes to airway remodeling and the deterioration of lung function. However, the initiating factor that links airway inflammation to remodeling is unknown. Thymic stromal lymphopoietin (TSLP), an epithelium-derived cytokine, can strongly activate lung dendritic cells (DCs) through the TSLP-TSLPR and OX40L-OX40 signalin...

IL-22 induces Reg3γ and inhibits allergic inflammation in house dust mite–induced asthma models

Previous studies have shown that IL-22, one of the Th17 cell-related cytokines, plays multiple roles in regulating allergic airway inflammation caused by antigen-specific Th2 cells; however, the underlying mechanism remains unclear. Here, we show that allergic airway inflammation and Th2 and Th17 cytokine production upon intratracheal administration of house dust mite (HDM) extract, a represent...

Sensitization by subcutaneous route is superior to intraperitoneal route in induction of asthma by house dust mite in a murine mode

OBJECTIVE To develop a new experimental model of chronic allergic pulmonary disease induced by house dust mite, with marked production of specific immunoglobulin E (IgE), eosinophilic inflammatory infiltrate in the airways and remodeling, comparing two different routes of sensitization. METHODS The protocol lasted 30 days. BALB/c mice were divided into six groups and were sensitized subcutane...
